Using tissue-specific RNA interference we tested in which tissues <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"60df84a8-b928-4d3c-9e4c-7c3f3b4006ae\">mlt-11</a> </i>activity was critical for molting and developmental progression. <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"d3cbe496-1c9a-459c-bc95-8242297d7292\">mlt-11</a> </i>knockdown using a seam cell-enriched RNAi strain produced similar phenotypes as knockdown throughout animals. In contrast, hypodermis-enriched knockdown caused no detectable phenotype. Understanding the mechanisms underlying molting can provide insight into oscillatory biological timers, epithelial biology, and is a source of potential targets to develop anthelminthics to control parasitic nematodes of medical and agricultural importance. Proteases and protease inhibitors play important and poorly understood roles in remodeling of the <i><a href=\"https://www.ncbi.nlm.nih.gov/Taxonomy/Browser/wwwtax.cgi?mode=Info&amp;id=6239\" id=\"90cfd6e3-35cd-4003-b2db-962ca22fba7f\">C. elegans</a> </i>aECM during molting.</p><p><i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"14b8833a-99a6-4603-a588-b5707753db0d\">mlt-11</a> </i>is a putative Kunitz family protease inhibitor that plays a critical role in <i>C.elegans </i>cuticle synthesis and molting <a href=\"https://www.zotero.org/google-docs/?dExfed\">(Frand et al., 2005; Ragle et al., 2026)</a>. It is a complex protein with 10 Kunitz domains and depending on which of these domains are deleted, phenotypes such as left rollers, right rollers, microblistering, or embryonic lethality <a href=\"https://www.zotero.org/google-docs/?2KWkNv\">(Ragle et al., 2026)</a>. Reduction of <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"66c664a5-3bd5-4343-ba20-a164b2ec5642\">mlt-11</a> </i>function results in aberrant protein localization in the aECM over both seam and hypodermal cells, and all three layers of the adult cuticle were affected (basal, medial, and cortical layers) <a href=\"https://www.zotero.org/google-docs/?OpHOMM\">(Ragle et al., 2026)</a>. Given that <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"3c3944d9-725a-4a8b-acd8-9f48a52f41ec\">mlt-11</a> </i>is expressed in both seam cells and in the hypodermal syncytium <a href=\"https://www.zotero.org/google-docs/?bGpNHT\">(Frand et al., 2005; Ragle et al., 2026)</a>, we turned to a tissue-specific RNAi approach  to test where <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"5ab6505a-a005-43ac-9b48-5c21d104e910\">mlt-11</a> </i>activity was needed to promote molting (Fig. 1A). <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"7dd0d2bd-b3b6-444c-bb4c-aa7bd34bb1d1\">mlt-11</a> </i>knockdown in a tissue-specific RNAi strain (<a id=\"5abca08b-5c0b-4247-afe5-16ea68034caa\">JDW371</a>) that restricts knockdown to seam, hypodermal, and intestinal cells <a href=\"https://www.zotero.org/google-docs/?U9m74f\">(Johnson et al., 2023)</a>, phenocopied <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"432b9346-f980-42c3-adfc-947b67fc2e88\">mlt-11</a> </i>RNAi in wildtype animals with respect to developmental delay and molting defects (Fig. 1B-D). <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"9351dcbc-64eb-4477-9082-1cfdfde7a0e3\">mlt-11</a> </i>RNAi in a hypodermal and seam cell-specific RNAi strain (<a href=\"http://www.wormbase.org/db/get?name=WBStrain00031246;class=Strain\" id=\"36a644ea-b42d-49bc-ae71-e78e4fa338da\">QK52</a>), produced less penetrant developmental delay and molting defects (Fig. 1B-D). Notably, <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"5c099095-e9fd-42ab-ac80-c9f221430cb1\">mlt-11</a> </i>RNAi in a hypodermal-specific RNAi strain (<a id=\"947392c8-e129-4318-9d06-bf25d18911b8\">JDW510</a>),produced no developmental delay or molting defects, suggesting <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"6d48d011-e131-4d72-b724-c4a2ca872d38\">mlt-11</a> </i>activity is necessary in seam cells and perhaps, in a limited capacity, in other hypodermal cells (Fig. 1B-D).</p><p>Despite robust hypodermal expression and localization <a href=\"https://www.zotero.org/google-docs/?56DjTf\">(Frand et al., 2005; Ragle et al., 2026)</a>, our tissue-specific RNAi suggests that the seam cells are an important site of <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"79254621-5a08-47f6-84d0-f1c11d8ef50c\">mlt-11</a> </i>expression (Fig. 1). We consistently saw the most severe RNAi phenotypes in tissue-specific RNAi strains using a seam cell enriched promoter and no phenotypes from a hypodermal/intestinal-specific RNAi strain (Fig. 6). Consistent with these data, we previously observed aberrant expression of the markers <a href=\"http://www.wormbase.org/db/get?name=WBGene00001069;class=Gene\" id=\"d1cdec55-3c97-472d-b6e0-11bfa144666f\">DPY-7</a>::GFP (furrows), <a href=\"http://www.wormbase.org/db/get?name=WBGene00001072;class=Gene\" id=\"075a7d85-14fa-4905-a796-25d015a7ce59\">DPY-10</a>::mScarlet (furrows), <a href=\"http://www.wormbase.org/db/get?name=WBGene00004397;class=Gene\" id=\"da35ac52-c61b-44d4-acde-a6f28bb8cb38\">ROL-6</a>::mNG (basal layer collagen), <a href=\"http://www.wormbase.org/db/get?name=WBGene00009983;class=Gene\" id=\"ad2ac73b-232a-487e-890a-42d1f9360334\">CUT-2</a>::mNG (cortical layer cuticlin), and <a href=\"http://www.wormbase.org/db/get?name=WBGene00000608;class=Gene\" id=\"aa809276-5c49-464e-bcc6-6e7ffa17d836\">COL-19</a>::mNG (basal layer collagen) over the seam cells as well as alae gaps following <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"865307a3-4a58-4035-9f13-d68db71217a0\">mlt-11</a> </i>inactivation <a href=\"https://www.zotero.org/google-docs/?b0vWpV\">(Ragle et al., 2026)</a>. The medial layer strut collagen <a href=\"http://www.wormbase.org/db/get?name=WBGene00000251;class=Gene\" id=\"9bfb622b-e181-4a89-9f63-7bb6a07f6781\">BLI-1</a> is normally excluded from the aECM over the seam cells but invades this area following <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"8846800e-33a7-4597-9700-660bca6d82d5\">mlt-11</a> </i>knockdown <a href=\"https://www.zotero.org/google-docs/?aaH0Ni\">(Ragle et al., 2026)</a>. These data were reminiscent of our work on <a href=\"http://www.wormbase.org/db/get?name=WBGene00003622;class=Gene\" id=\"9f1f6c6f-1478-4cb6-9c64-4094194d90a6\">NHR-23</a> as depletion of this factor causes aberrant aECM formation over the seam cells and <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00003622;class=Gene\" id=\"e73ae365-de6c-472b-808b-804e3910e4a2\">nhr-23</a> </i>activity also was necessary in the seam cells for developmental progression <a href=\"https://www.zotero.org/google-docs/?GJuYCT\">(Johnson et al., 2023)</a>. Seam cells are responsible for the production of alae <a href=\"https://www.zotero.org/google-docs/?zqX68U\">(Singh &amp; Sulston, 1978)</a>, so it is unclear why inactivation of <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00003622;class=Gene\" id=\"7319c111-065d-4d3e-8176-0633620deed6\">nhr-23</a> </i>or <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"4919d6aa-cb49-46bc-a8c0-32888bc71b08\">mlt-11</a> </i>in the seam cells has such a potent effect on developmental progression and molting given that the hypodermis secretes the bulk of the cuticle. Understanding the mechanisms underlying molting can provide insight into oscillatory biological timers, epithelial biology, and is a source of potential targets to develop anthelminthics to control parasitic nematodes of medical and agricultural importance. Proteases and protease inhibitors play important and poorly understood roles in remodeling of the <i><a href=\"https://www.ncbi.nlm.nih.gov/Taxonomy/Browser/wwwtax.cgi?mode=Info&amp;id=6239\" id=\"90cfd6e3-35cd-4003-b2db-962ca22fba7f\">C. elegans</a> </i>aECM during molting.</p><p><i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"14b8833a-99a6-4603-a588-b5707753db0d\">mlt-11</a> </i>is a putative Kunitz family protease inhibitor that plays a critical role in <i>C.elegans </i>cuticle synthesis and molting <a href=\"https://www.zotero.org/google-docs/?dExfed\">(Frand et al., 2005; Ragle et al., 2026)</a>. It is a complex protein with 10 Kunitz domains and depending on which of these domains are deleted, phenotypes such as left rollers, right rollers, microblistering, or embryonic lethality <a href=\"https://www.zotero.org/google-docs/?2KWkNv\">(Ragle et al., 2026)</a>. Reduction of <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"66c664a5-3bd5-4343-ba20-a164b2ec5642\">mlt-11</a> </i>function results in aberrant protein localization in the aECM over both seam and hypodermal cells, and all three layers of the adult cuticle were affected (basal, medial, and cortical layers) <a href=\"https://www.zotero.org/google-docs/?OpHOMM\">(Ragle et al., 2026)</a>. Given that <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"3c3944d9-725a-4a8b-acd8-9f48a52f41ec\">mlt-11</a> </i>is expressed in both seam cells and in the hypodermal syncytium <a href=\"https://www.zotero.org/google-docs/?bGpNHT\">(Frand et al., 2005; Ragle et al., 2026)</a>, we turned to a tissue-specific RNAi approach  to test where <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"5ab6505a-a005-43ac-9b48-5c21d104e910\">mlt-11</a> </i>activity was needed to promote molting (Fig. 1A). <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"7dd0d2bd-b3b6-444c-bb4c-aa7bd34bb1d1\">mlt-11</a> </i>knockdown in a tissue-specific RNAi strain (<a href=\"https://wormbase.org/species/c_elegans/strain/WBStrain00064005\" id=\"c989d680-ab96-4052-9731-77479790ac15\">JDW371</a>) that restricts knockdown to seam, hypodermal, and intestinal cells <a href=\"https://www.zotero.org/google-docs/?U9m74f\">(Johnson et al., 2023)</a>, phenocopied <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"432b9346-f980-42c3-adfc-947b67fc2e88\">mlt-11</a> </i>RNAi in wild-type animals with respect to developmental delay and molting defects (Fig. 1B-D). <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"9351dcbc-64eb-4477-9082-1cfdfde7a0e3\">mlt-11</a> </i>RNAi in a hypodermal and seam cell-specific RNAi strain (<a href=\"http://www.wormbase.org/db/get?name=WBStrain00031246;class=Strain\" id=\"36a644ea-b42d-49bc-ae71-e78e4fa338da\">QK52</a>), produced less penetrant developmental delay and molting defects (Fig. 1B-D). Notably, <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"5c099095-e9fd-42ab-ac80-c9f221430cb1\">mlt-11</a> </i>RNAi in a hypodermal-specific RNAi strain (<a href=\"https://wormbase.org/species/c_elegans/strain/WBStrain00064006\" id=\"35ae6d56-342f-481e-8915-67dd9f1035d9\">JDW510</a>),produced no developmental delay or molting defects, suggesting <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"6d48d011-e131-4d72-b724-c4a2ca872d38\">mlt-11</a> </i>activity is necessary in seam cells and perhaps, in a limited capacity, in other hypodermal cells (Fig. 1B-D).</p><p>Despite robust hypodermal expression and localization <a href=\"https://www.zotero.org/google-docs/?56DjTf\">(Frand et al., 2005; Ragle et al., 2026)</a>, our tissue-specific RNAi suggests that the seam cells are an important site of <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"79254621-5a08-47f6-84d0-f1c11d8ef50c\">mlt-11</a> </i>expression (Fig. 1). We consistently saw the most severe RNAi phenotypes in tissue-specific RNAi strains using a seam cell enriched promoter and no phenotypes from a hypodermal/intestinal-specific RNAi strain (Fig. 1B-D). Consistent with these data, we previously observed aberrant expression of the markers <a href=\"http://www.wormbase.org/db/get?name=WBGene00001069;class=Gene\" id=\"d1cdec55-3c97-472d-b6e0-11bfa144666f\">DPY-7</a>::GFP (furrows), <a href=\"http://www.wormbase.org/db/get?name=WBGene00001072;class=Gene\" id=\"075a7d85-14fa-4905-a796-25d015a7ce59\">DPY-10</a>::mScarlet (furrows), <a href=\"http://www.wormbase.org/db/get?name=WBGene00004397;class=Gene\" id=\"da35ac52-c61b-44d4-acde-a6f28bb8cb38\">ROL-6</a>::mNG (basal layer collagen), <a href=\"http://www.wormbase.org/db/get?name=WBGene00009983;class=Gene\" id=\"ad2ac73b-232a-487e-890a-42d1f9360334\">CUT-2</a>::mNG (cortical layer cuticlin), and <a href=\"http://www.wormbase.org/db/get?name=WBGene00000608;class=Gene\" id=\"aa809276-5c49-464e-bcc6-6e7ffa17d836\">COL-19</a>::mNG (basal layer collagen) over the seam cells as well as alae gaps following <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"865307a3-4a58-4035-9f13-d68db71217a0\">mlt-11</a> </i>inactivation <a href=\"https://www.zotero.org/google-docs/?b0vWpV\">(Ragle et al., 2026)</a>. The medial layer strut collagen <a href=\"http://www.wormbase.org/db/get?name=WBGene00000251;class=Gene\" id=\"9bfb622b-e181-4a89-9f63-7bb6a07f6781\">BLI-1</a> is normally excluded from the aECM over the seam cells but invades this area following <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"8846800e-33a7-4597-9700-660bca6d82d5\">mlt-11</a> </i>knockdown <a href=\"https://www.zotero.org/google-docs/?aaH0Ni\">(Ragle et al., 2026)</a>. These data were reminiscent of our work on <a href=\"http://www.wormbase.org/db/get?name=WBGene00003622;class=Gene\" id=\"9f1f6c6f-1478-4cb6-9c64-4094194d90a6\">NHR-23</a> as depletion of this factor causes aberrant aECM formation over the seam cells and <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00003622;class=Gene\" id=\"e73ae365-de6c-472b-808b-804e3910e4a2\">nhr-23</a> </i>activity also was necessary in the seam cells for developmental progression <a href=\"https://www.zotero.org/google-docs/?GJuYCT\">(Johnson et al., 2023)</a>. Seam cells are responsible for the production of alae <a href=\"https://www.zotero.org/google-docs/?zqX68U\">(Singh &amp; Sulston, 1978)</a>, so it is unclear why inactivation of <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00003622;class=Gene\" id=\"7319c111-065d-4d3e-8176-0633620deed6\">nhr-23</a> </i>or <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"4919d6aa-cb49-46bc-a8c0-32888bc71b08\">mlt-11</a> </i>in the seam cells has such a potent effect on developmental progression and molting given that the hypodermis secretes the bulk of the cuticle. <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"8846800e-33a7-4597-9700-660bca6d82d5\">mlt-11</a></i> inactivation has been shown to cause reduced body length and width, impaired cuticle barrier function, and aberrant aECM protein localization <a href=\"https://www.zotero.org/google-docs/?aaH0Ni\">(Ragle et al., 2026)</a>. Understanding the mechanisms underlying molting can provide insight into oscillatory biological timers, epithelial biology, and is a source of potential targets to develop anthelminthics to control parasitic nematodes of medical and agricultural importance. Proteases and protease inhibitors play important and poorly understood roles in remodeling of the <i><a href=\"https://www.ncbi.nlm.nih.gov/Taxonomy/Browser/wwwtax.cgi?mode=Info&amp;id=6239\" id=\"90cfd6e3-35cd-4003-b2db-962ca22fba7f\">C. elegans</a> </i>aECM during molting.</p><p><i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"14b8833a-99a6-4603-a588-b5707753db0d\">mlt-11</a> </i>is a putative Kunitz family protease inhibitor that plays a critical role in <i>C.elegans </i>cuticle synthesis and molting (Frand et al., 2005; Ragle et al., 2026). It is a complex protein with 10 Kunitz domains and depending on which of these domains are deleted, phenotypes such as left rollers, right rollers, microblistering, or embryonic lethality are observed (Ragle et al., 2026). Reduction of <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"66c664a5-3bd5-4343-ba20-a164b2ec5642\">mlt-11</a> </i>function results in aberrant protein localization in the aECM over both seam and hypodermal cells, and all three layers of the adult cuticle were affected (basal, medial, and cortical layers) (Ragle et al., 2026). Given that <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"3c3944d9-725a-4a8b-acd8-9f48a52f41ec\">mlt-11</a> </i>is expressed in both seam cells and in the hypodermal syncytium (Frand et al., 2005; Ragle et al., 2026), we turned to a tissue-specific RNAi approach  to test where <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"5ab6505a-a005-43ac-9b48-5c21d104e910\">mlt-11</a> </i>activity was needed to promote molting (Fig. 1A). <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"7dd0d2bd-b3b6-444c-bb4c-aa7bd34bb1d1\">mlt-11</a> </i>knockdown in a tissue-specific RNAi strain (<a href=\"https://wormbase.org/species/c_elegans/strain/WBStrain00064005\" id=\"c989d680-ab96-4052-9731-77479790ac15\">JDW371</a>) that restricts knockdown to seam, hypodermal, and intestinal cells (Johnson et al., 2023), phenocopied <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"432b9346-f980-42c3-adfc-947b67fc2e88\">mlt-11</a> </i>RNAi in wild-type animals with respect to developmental delay and molting defects (Fig. 1B-D). <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"9351dcbc-64eb-4477-9082-1cfdfde7a0e3\">mlt-11</a> </i>RNAi in a hypodermal and seam cell-specific RNAi strain (<a href=\"http://www.wormbase.org/db/get?name=WBStrain00031246;class=Strain\" id=\"36a644ea-b42d-49bc-ae71-e78e4fa338da\">QK52</a>), produced less penetrant developmental delay and molting defects (Fig. 1B-D). Notably, <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"5c099095-e9fd-42ab-ac80-c9f221430cb1\">mlt-11</a> </i>RNAi in a hypodermal and intestinal-specific RNAi strain (<a href=\"https://wormbase.org/species/c_elegans/strain/WBStrain00064006\" id=\"35ae6d56-342f-481e-8915-67dd9f1035d9\">JDW510</a>), produced no developmental delay or molting defects, suggesting <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"6d48d011-e131-4d72-b724-c4a2ca872d38\">mlt-11</a> </i>activity is necessary in seam cells and perhaps, in a limited capacity, in other hypodermal cells (Fig. 1B-D).</p><p>Despite robust hypodermal expression and localization (Frand et al., 2005; Ragle et al., 2026), our tissue-specific RNAi suggests that the seam cells are an important site of <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"79254621-5a08-47f6-84d0-f1c11d8ef50c\">mlt-11</a> </i>expression (Fig. 1). We consistently saw the most severe RNAi phenotypes in tissue-specific RNAi strains using a seam cell enriched promoter and no phenotypes from a hypodermal/intestinal-specific RNAi strain (Fig. 1B-D). Consistent with these data, we previously observed aberrant expression of the markers <a href=\"http://www.wormbase.org/db/get?name=WBGene00001069;class=Gene\" id=\"d1cdec55-3c97-472d-b6e0-11bfa144666f\">DPY-7</a>::GFP (furrows), <a href=\"http://www.wormbase.org/db/get?name=WBGene00001072;class=Gene\" id=\"075a7d85-14fa-4905-a796-25d015a7ce59\">DPY-10</a>::mScarlet (furrows), <a href=\"http://www.wormbase.org/db/get?name=WBGene00004397;class=Gene\" id=\"da35ac52-c61b-44d4-acde-a6f28bb8cb38\">ROL-6</a>::mNG (basal layer collagen), <a href=\"http://www.wormbase.org/db/get?name=WBGene00009983;class=Gene\" id=\"ad2ac73b-232a-487e-890a-42d1f9360334\">CUT-2</a>::mNG (cortical layer cuticlin), and <a href=\"http://www.wormbase.org/db/get?name=WBGene00000608;class=Gene\" id=\"aa809276-5c49-464e-bcc6-6e7ffa17d836\">COL-19</a>::mNG (basal layer collagen) over the seam cells as well as alae gaps following <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"865307a3-4a58-4035-9f13-d68db71217a0\">mlt-11</a> </i>inactivation (Ragle et al., 2026). The medial layer strut collagen <a href=\"http://www.wormbase.org/db/get?name=WBGene00000251;class=Gene\" id=\"9bfb622b-e181-4a89-9f63-7bb6a07f6781\">BLI-1</a> is normally excluded from the aECM over the seam cells but invades this area following <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"8846800e-33a7-4597-9700-660bca6d82d5\">mlt-11</a> </i>knockdown (Ragle et al., 2026). These data were reminiscent of our work on <a href=\"http://www.wormbase.org/db/get?name=WBGene00003622;class=Gene\" id=\"9f1f6c6f-1478-4cb6-9c64-4094194d90a6\">NHR-23</a> as depletion of this factor causes aberrant aECM formation over the seam cells and <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00003622;class=Gene\" id=\"e73ae365-de6c-472b-808b-804e3910e4a2\">nhr-23</a> </i>activity also was necessary in the seam cells for developmental progression (Johnson et al., 2023). Seam cells are responsible for the production of alae (Singh &amp; Sulston, 1978), so it is unclear why inactivation of <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00003622;class=Gene\" id=\"7319c111-065d-4d3e-8176-0633620deed6\">nhr-23</a> </i>or <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"4919d6aa-cb49-46bc-a8c0-32888bc71b08\">mlt-11</a> </i>in the seam cells has such a potent effect on developmental progression and molting given that the hypodermis secretes the bulk of the cuticle. <i><a href=\"http://www.wormbase.org/db/get?name=WBGene00012186;class=Gene\" id=\"8846800e-33a7-4597-9700-660bca6d82d5\">mlt-11</a></i> inactivation has been shown to cause reduced body length and width, impaired cuticle barrier function, and aberrant aECM protein localization (Ragle et al., 2026).
